Biomimetic material A recent study by Wu, Zheng et al., synthesized a self-assembled IKVAV peptide nanofiber and tested its effect on the adhesion of neuron-like pc12 cells. Early cell adhesion is very important for preventing cell degeneration; the longer cells are suspended in culture, the more likely they are to degenerate. The purpose was to develop a biomaterial with good cell adherence and bioactivity with IKVAV, which is able to inhibit differentiation and adhesion of glial cells in addition to promoting neuronal cell adhesion and differentiation. The IKVAV peptide domain is on the surface of the nanofibers so that it is exposed and accessible for promoting cell contact interactions. The IKVAV nanofibers promoted stronger cell adherence than the electrostatic attraction induced by poly-L-lysine, and cell adherence increased with increasing density of IKVAV until the saturation point was reached. IKVAV does not exhibit time dependent effects because the adherence was shown to be the same at 1 hour and at 3 hours.
Biomimetic material Many studies utilize laminin-1 when designing a biomimetic material. Laminin is a component of the extracellular matrix that is able to promote neuron attachment and differentiation, in addition to axon growth guidance. Its primary functional site for bioactivity is its core protein domain isoleucine-lysine-valine-alanine-valine (IKVAV), which is located in the α-1 chain of laminin.
Fibrin scaffold Fibrin gels can be enriched by addition of other extracellular matrix (ECM) components such as fibronectin, vitronectin, laminin and collagen. These can be linked covalently to fibrin scaffold by reactions catalyzed by transglutaminase. Laminin originated substrate amino acid sequences for transglutaminase can be IKVAV, YIGSR or RNIAEIIKDI. Collagen originated sequence is DGEA and many other ECM protein originated RGD sequence can be given as other examples. Heparin binding sequences KβAFAKLAARLYRKA, RβAFARLAARLYRRA, KHKGRDVILKKDVR, YKKIIKKL are from antithrombin III, modified antithrombin III, neural cell adhesion molecule and platelet factor 4, respectively. Heparin-binding growth factors can be attached to heparin binding domains via heparin. As a result, a reservoir can be provided instead of passive diffusion by liberation of growth factors in extended time. Acidic and basic fibroblast growth factor, neurotrophin 3, transforming growth factor beta 1, transforming growth factor beta 2, nerve growth factor, brain derived neurotrophic factor can be given as examples for such growth factors.
Surface modification of biomaterials with proteins Peripheral nervous system damage is typically treated by an autograft of nerve tissue to bridge a severed gap. This treatment requires successful regeneration of neural tissue; axons must grow from the proximal stump without interference in order to make a connection with the distal stump. Neural guidance channels (NGC), have been designed as a conduit for growth of new axons and the differentiation and morphogenesis of these tissues is affect by interaction between neural cells and the surrounding ECM. Studies of laminin have shown the protein to be an important ECM protein in the attachment of neural cells. The penta-peptide YIGSR and IKVAV, which are important sequences in laminin, have been shown to increase attachment of neural cells with the ability to control the spatial organization of the cells.
Peptide amphiphile The modular nature of the chemistry allows the tuning of both the mechanical properties and bioactivities of the resulting self-assembled fibers and gels. Bioactive sequences can be used to bind growth factors to localize and present them at high densities to cells, or to directly mimic the function of endogenous biomolecules. Epitopes mimicking the adhesive RGD loop in fibronectin, the IKVAV sequence in laminin and a consensus sequence to bind heparin sulfate are just a few of the large library of sequences that have been synthesized. These molecules and the materials made from them have been shown to be effective in promoting cell adhesion, wound healing, mineralization of bone, differentiation of cells and even recovery of function after spinal cord injury in mice.
